Former Red Luis Suarez gave Barcelona the lead in the first half, before Lionel Messi scored two second half goals.
The Argentina striker is likely to have put his team beyond reach with a stunning free kick 8 minutes from time.
Messi bent the ball around the wall from 25 yards and into the top corner for his second goal of the game and his 600th career goal for Barcelona.
The second leg takes place at Anfield next Tuesday.
